It's been about four years since the events of Guardians of the Galaxy Vol. 2, and the gang is even more of a family now than ever. Groot has even reached adolescence.
The new version of the Milano, the Benatar, is racing towards a distress signal, in hopes of both finding people to save and also maybe sorta getting something in return for it. Thor suddenly crashes onto their windshield and he's retrieved and revived back into consciousness. Thanks to Thor's information, group discovers that Thanos is after the Infinity Stones again and has now acquired two of them, and destroyed Xandar the previous week. Thor has deduced where Thanos will go next. They decide to split into two groups--Rocket and Groot will go with Thor to Nidavellir to make a Thanos-killing weapon, and Peter, Gamora, Mantis, and Drax will go to Knowhere to stop Thanos from getting the Reality Stone.
En route to Knowhere, Gamora makes Peter promise, on his mother, that he will kill Gamora if Thanos somehow gets to her. It's a heartbreaking promise, but Peter agrees to it, hoping it will never come to that. They kiss, but unfortunately Drax ruins the moment.
Knowhere seems deserted, and the gang finds Thanos interrogating the Collector. Nobody listens to Peter when he tries to guide them on how to approach carefully, Drax nearly runs ahead but Mantis stops him--unfortunately, his body crashing down makes a loud noise. Peter tries to tell Gamora which way to go but she doesn't listen, and attacks and seemingly kills Thanos. Unfortunately it's only an illusion and he captures her, using the Reality Stone to split Drax up and turn Mantis into ribbons. Peter approaches, meaning to shoot Thanos, but Gamora insists he shoots her instead. Peter's absolutely devastated, he can't believe this is actually happening, but he will keep his promise to Gamora even as Thanos teases them both by moving her closer. The two confess their love for each other before he fires, but it turns out to be much worse--his gun shoots bubbles instead and disappears, Thanos commenting that he likes Peter, since he was able to follow through with that decision to shoot. He disappears with Gamora, and without the Reality Stone in proximity, Drax and Mantis return to normal Peter picks up Gamora's sword in shock.
Mantis receives a message from Nebula to meet her on Titan, where they'll be expecting Thanos to show up next. On Titan, Peter and his remaining crew attack the inhabitants of a newly crashed ship, which are actually Peter Parker, Dr. Stephen Strange, and Tony Stark. There's a tense stand-off until Tony figures out that Peter is from earth, and they all try to make a plan of attack. Stephen looks into the future at this point as well.
Once Thanos arrives on Titan, their plan goes surprisingly well and they manage to pin him down long enough to start to get the gauntlet off. It's also Peter's plan, which he's pretty smug about, and he tries to get Thanos, under Mantis' spell, to reveal to him where Gamora is. Unfortunately this makes things so much worse--between what Mantis says about him mourning, and Nebula revealing that he's got the Soul Stone and Gamora's not with him, implies that Gamora is dead. Peter absolutely snaps in two, and demands to know if it's true--when Thanos confirms it, he loses it and starts hitting him wildly in anguished rage, having to be held back by Tony. This wakes Thanos up and he's able to break loose even though they almost got the Gauntlet off.
They continue to battle him--Peter gets knocked out by a blast from the power stone and is saved by Spider-Man when Thanos brings a moon down on them. Peter tries to make one last attack after Stephen gives the Time Stone to Thanos, but it's too late--he disappears, traveling to earth, and a distraught Peter asks, "Did we just lose?"
Not long after that when they all regroup, Mantis notices something is happening. She suddenly disintegrates in a puff of dust as Peter's holding onto her. He looks over at Drax, who is also disintegrating and frightened--and he also disappears in a puff of dust. It's happening, Thanos has done the snap, and Peter's just lost nearly everyone. Tony tries to keep him from panicking by saying, "steady Quill," but it's too late. Peter too disintegrates into dust after a defeated, "Oh, man."
